药品注册研发SMP or SOP——本SOP中惨杂了太多个人观点，请自行判断，若引人误入歧途，概不负责，特此声明。1. 目的：建立药品注册研发的一般规程，保证药品注册申报审评过程的顺利进行，缩短研发、审评及审批的时限，最大限度降低研发风险，达到“真做药，做真药”之目的。2. 依据：现行《药品注册管理办法》及附件，相关的药品研究指导原则和官方的现场核查要求。3. 范围：适用于shyflysky药业研发部中药、天然药物、化学药品的注册管理，包括新药及已有国家标准的药品。4. 职责：研发部主管负责新药及已有国家标准的药品的立项，同时监督、检查项目进展，上报前最后审核申报资料、原始纪录；各课题组长及项目负责人承担课题项目，负责完成立项后的各项申报前研究工作（不包括外协工作），各课题组长负责监督、检查本组项目进展，项目结束前审核资料和纪录，连同项目清单转交给研发部主管审核。5.
程序：5.1 起草项目建议书研发部主管负责新药及已有国家标准的药品的立项，起草项目建议书。建议书中至少涵盖以下内容：（a）
国内外上市情况（b）
专利权属状态、行政保护及法规保护情形（c）
SFDA受理申报情况（d）
质量标准、原料（药材）来源、处方组成及说明书（e）
市场成本分析（f）
立项风险（g）
临床试验资料、文献资料支持5.2 立项研发部主管依据项目建议书向公司技术管理委员会提出立项申请或中止开发项目建议。公司技术管理委员会对项目建议书进行审议，形成决议案，由总裁批准立项。5.3 项目开展研发部主管安排项目给各课题组，课题组长安排项目负责人选，填写项目设计方案草稿和项目计划任务书。（a）课题组长或项目负责人一周内将设计方案草稿及计划书书面递交给研发主管，同时启动原辅料采购和文献资料查阅工作。（b）原辅料的购买需要厂家提供证明性文件包括但不限于营业执照、生产许可证、检验报告、质量标准、批准证明文件、供货发票等，原料药还应与厂家签署供货协议或购销合同，这些证明性文件各项目负责人手中至少留有复印件，这些证明性文件放置在一个档案袋中，档案袋上注明项目或代号，同时在各复印件背面注明原件的存放处。（c）原辅料到货后，及时联系质控部进行进厂检验，通常情况下，原料药和药材应当全检，而辅料和包材只需在厂家报告书的基础上部分检验，并将检验记录和出入库单备案，以备日后核查用。另一方面，在文献资料的基础上，开始处方工艺研究，同时按照《试验室管理规范》和核查要求书写工艺研究记录。《试验室管理规范》见附件一。（d）按照既定的处方工艺，试制小试样品（批产量500~10000粒、片、袋等），进行影响因素试验（中药可以不做），进行初步的质量研究，这部分的研究记录可以在公司内部记录上书写（因为国家局要求质量研究用样品应具一定规模），初步的质量研究工作为日后的以制订质量标准为目的的质量研究做铺垫。（e）中试放大：应在GMP车间或符合GMP要求的试验室中进行，按照GMP要求编写批生产记录，批生产记录中应有检验报告书。关于申报生产样品的试制量，出台了一个所谓“最大量\半数量和最小量”的265号文件，实施起来难度巨大，而且与注册管理办法中的“连续3个生产批号的样品”有一定出入。（f）在初步的质量研究基础之上，并参阅相关标准和文献，对中试样品进行质量研究，研究项目主要有[性状]、[鉴别]、[检查]、[含量测定]等，各制剂参阅中国药典2005年版附录的制剂通则。编写质量研究记录，记录中的各检验项目应包括但不限于仪器名称、仪器型号、试剂、试验方法等，薄层照片、紫外扫描图谱、高效液相色谱图等附在相应的项目之后，色谱图应包含进样时间、流速等相关信息，所有TLC照片、色谱图等应有电子版备份。大型仪器应有仪器使用记录。质量研究过程中的各项内容应符合逻辑，如方法的选择、检测波长的选择、流动相的选择、溶出介质的选择、展开剂的选择等应阐明依据。（g）试制样品结束后不久（一周内），开始安排加速试验和长期试验，无论申报生产还是申报临床申请，加速试验均进行6个月，长期试验至少6个月，同为3批样品（注：关于质量研究和稳定性研究的时间安排问题，争论已久，如果3批质量用样品同时也是稳定性样品，建议生产结束后不久开始加速长期试验）。（h）根据质量研究结果，参考相关资料，按照中国药典2005年格式起草质量标准。同时，如果3批质量用样品同时也是稳定性样品，质量研究结果正是加速试验0月和长期试验0月的检验结果，唯一有所不同的是，0月是全检，而稳定性其他各取样点部分检验。按照拟定的质量标准对加速1月、2月、3月、6月及长期试验（中药：1月、2月、3月、6月、12月、18月；化药：3月、6月、9月、12月、18月、24月）的样品进行检验，并书写加速试验记录和
